- Fan Nickname: Amazing, 2099, Noir, and Ultimate to differentiate between the Spider-Men.
- Hey, It's That Voice!: Several members of the cast included voice actors from past incarnations of Spider-Man and Marvel lore, including the Spider-Men themselves. Every Spider-Man in the game is voiced by someone who has voiced Spider-Man in the past. Amazing Spider-Man is voiced by Neil Patrick Harris, Noir is played by Christopher Daniel Barnes, Spider-Man 2099 is voiced by
BumblebeeDan Gilvezan, and Ultimate Spider-Man is voiced by Josh Keaton. Keaton also does the voice of Amazing in the DS version.- While not one of the Spider-Men, James Arnold Taylor lends his voice to a few hostages in Noir.
- Nolan North voices Ultimate Deadpool. John Dimaggio voices Noir Hammerhead. Steven Blum voices Hobgoblin 2099 (well, he voiced a Goblin before) and he'll also voice Noir Vulture; on the DS, Blum did the voices for the 2099 versions of Silvermane and Vulture.
- Jim Cummings (voiced Shocker in the 90's cartoon) is voicing Kraven and Noir Goblin. In the DS, he also voiced Tinkerer and Noir Boomerang.
- John Kassir (Deadpool in two video game series) is voicing Scorpion 2099 and some of Ultimate Deadpool's fanboy mooks.
- Fred Tatasciore is voicing Ultimate Carnage.
- David Kaye (Professor Xavier, Apocalypse, Electro, Iron Man, Captain America, Nick Fury) is Mysterio. And Kaye is no stranger to Large Hams--hell, Mysterio has Beast Wars Megatron's voice!
- Thomas F. Wilson (Stan Carter, and yes, Biff Tannen) is Electro.
- Jennifer Hale is once again Silver Sable (and that's not the only Spidey character she's voiced in the past). She also does the voice of Noir Calypso.
- Tara Strong lending her wonderful talents now to voice Lady Doc Ock and also being no stranger to Spider-Man.
- Madame Web is voiced by Susanne Blakeslee, which definetly will make some Kingdom Hearts fans raise an eyebrow whenever she gives any advice. After all, one must be cautious before taking orders from the MISTRESS OF ALL EVIL!
- Additional voices include the aforementioned James Arnold Taylor (Mysterio in the Spider-Man 2 tie-in game and Marvel Ultimate Alliance, Electro in Ultimate Spider-Man, Spidey himself in Spider-Man: Battle for New York and Spider-Man: Friend or Foe, and Harry Osborn in The Spectacular Spider-Man), Keith Szarabajka (the right-hand man of the Kingpin in an episode of Spider-Man: The New Animated Series, Wolverine in Ultimate Spider-Man and Venom in Spider-Man: Web of Shadows), Gregg Berger (Kraven and Mysterio in Spider-Man: The Animated Series and Kingpin in Spider-Man: Web of Shadows), and Nick Jameson (Morbius and Richard Fisk in Spider-Man: The Animated Series)
- I Knew It!: The aforementioned report of Neil Patrick Harris as Amazing Spider-Man. Before the announcement of his involvment, people speculated who was voicing Amazing Spidey. The list of suspects included Rino Romano (Spider-Man Unlimited and Activision's first Spider-Man games) and James Arnold Taylor (Spider-Man: Friend or Foe). When the Juggernaut teaser came out and had a few lines from Amazing Spidey, people pretty much figured out Harris was the voice. The report of his invovlment was pretty much just confirmation by the point it was made.
- When the image of the Cosmic Spider-Man costumes leaked between the initial anouncement & the 2099 reveal, most people immediately figured out that one of the two unanounced Spider-Men was Spider-Man 2099. On that same note, a smaller amount guessed that the last Spider-Man was wearing the Black suit & a smaller portion guessed that it was Ultimate Spider-Man (Far more had guessed that it was the Spidey from Web Of Shadows, following a Black Suit ending).
