- Actor Allusion: A subtle one: The Joker attempts to force Mr. Freeze to develop a cure for him by holding his wife hostage. The Joker is voiced by Mark Hamill, whose first voice in Batman: The Animated Series was Ferris Boyle, the guy responsible for Mr. Freeze, and Hamill's performance as him also landed him with his voicing The Joker in that series.
- Hey, It's That Voice!: Kevin Conroy and Mark Hamill are again reprising the roles of Batman and the Joker. Likewise, Tom Kane, Kimberly Brooks, Wally Wingert, Tasia Valenza, Fred Tatasciore, Danny Jacobs, and James Horan reprise their respective roles from the first game as Quincy Sharp, Oracle, the Riddler, Poison Ivy, Bane, Zsasz, and Jack Ryder. Though Tom doesn't reprise the role of Commissioner Gordon and Fred gets double duty as the voice of Solomon Grundy.
- Kevin Conroy also plays Thomas Elliot.
- Kate Beckett is Talia al Ghul.

- David Kaye takes over the role of Commissioner Gordon from Tom Kane.

- Nolan North plays Penguin, but puts on a cockney accent for the role. Makes it hard to believe that Deadpool/Superboy/Nathan Drake is really him. He is also Black Mask.
- Hugo Strange is voiced by Corey Burton, once again emulating Christopher Lee.
- Steve Blum lends his talent, of course. But his performance as a random thug in the beginning of the game will have you blinking and wondering, "When did Oghren get so tall?"
- Mad Hatter wants both Batman's cowl (and his belt) and the crest on Birdman's helmet.
- He was also Kirk Langstrom aka Man-Bat as well as Doctor Octopus.
- Khary Payton a.k.a. Cyborg and Aqualad is Azrael.
- Kari Wahlgren is Vicki Vale, which begs the question: When did Raine Sage become a news reporter?
- Rick D. Wasserman (Thor) is Clayface.
- Chris Cox (Captain Atom, Hawkeye) is Deadshot.
- Killer Croc is now voiced by Dee Bradley Baker, who is certainly no stranger to voicing reptiles.
