- Ezio Auditore is Gilgamesh and Domon Kasshu.
- Mario Auditore is Solid Snake and Zero's Rider.
- Giovanni Auditore is Madara Uchiha.
- Federico Auditore Tetsuya Onodera.
- Cesare Borgia is Archer and Relius Clover.
- Lucrezia Borgia is Saber and Leina.
- Cristina Vespucci is Haruhi Suzumiya and Konata Izumi.
- Leonardo da Vinci is Tekkaman Blade and Kyosuke Nanbu.
- Micheletto Corella is Ali-al Saachez.
- Caterina Sforza is Nunnally Lamperouge and Nora.

- In a rare in-series example, the voice of the Damascus Bureau rafiq in AC1 returns as the voice of Leonardo da Vinci in Assassin's Creed II. Out-of-series, he's also Dom.
- Fred Tatasciore plays Uncle Mario, but he also played Abu'l Nuqoud in the first game.
- Veronica Mars didn't join the FBI after season 3... she joined the Assassins!
- Carlo Grimaldi is also a sleazy alien planet salesman.
- I Knew It!: Fans speculating that Maria would be Altaïr's Love Interest were right.
- Relationship Voice Actor: The aforementioned Japanese version has many cast members that worked together not only in Fate/stay night, but in other titles:
- Ezio, Cesare and Lucrezia were Gilgamesh, Archer and Saber respectively. Special mention towards the relation between Ezio and Lucrezia since their roles are switched out from their roles in Fate.
- Also Lucrezia and Cristina Vespucci were Leina and Nanael, except they don't interact this time. And, continuing with Lucrezia's VA and her relationship with the rest of the cast, Lucrezia and Leonardo da Vinci were Dante and Beatrice in the Japanese version, to the grade all those four characters their voice actors voiced are Italians in both games.
- Ezio, Leonardo da Vinci and Micheletto Corella were Ishida Mitsunari, Katakura Kojuro and Matsunaga Hisahide. And just like with Ezio and Lucrezia, Ezio and Leonardo's roles are switched out this time from Sengoku Basara.
- Science Marches On: You can occasionally hear the doctors advocating medicines based on lead.
